Lab facilities

Separation techniques

  • FPLC (NGC Quest 10 Plus Chromatography System, BioRad) – setup for chromatographic purification of proteins, detection of up to four wavelengths simultaneously, conductivity detection, fraction collector (BioFrac).
  • HPLC (Perkin Elmer Flexar) – two detectors: diode array (190-700 nm) and fluorescent (excitation 200-850 nm, emission 250-900 nm), a gradient pump capable of creating a gradient composition from four components
  • Electrophoresis – protein vertical gel electrophoresis, western blotting, DNA horizontal gel electrophoresis (BioRad: Mini-PROTEAN Tetra Vertical Electrophoresis Cell, Mini Trans-Blot® Cell, Mini-Sub Cell GT Cell).

Absorption and emission spectroscopy

  • UV-Vis spectrophotometers (Perkin Elmer: Lambda 35, Lambda 265 PDA) – temperature control, repeat scan mode, kinetic mode.
  • Spectrofluorometer (Perkin Elmer LS 55) - temperature control, repeat scan mode, kinetic mode.
  • Stopped-flow spectrofluorometer (Applied Photophysics SX20) - UV-Vis and fluorescence detection, pre-mix system, reactions are following in solution over timescales in the range of one millisecond to hundreds of seconds.
  • Fluorolog-3 Horiba Jobin Yvon – steady-state and time-resolved spectrofluorometer, Time-Correlated Single Photon Counting mode, NanoLed Diodes sets (265, 350, 465 nm).

Special equipment

  • ICP-MS (NexION 2000C, Perkin Elmer) – three quadrupoles, three gas channels (He, O2, CH4), and three modes of operation (standard, KED, DRC).
  • Dianthus NT.23Pico (Nanotemper) – plate-based (384 well plate) affinity screening platform for evaluation protein-ligand interactions, it relies on  Temperature Related Intensity Change (TRIC).
  • Prometheus Panta (Nanotemper) – it uses label-free nanoDSF (nano differential scanning fluorimetry) for characterization of thermal unfolding or chemical denaturation, back-reflection for aggregate formation, DLS (differential light scattering) for protein size determination, and SLS (static light scattering) for molecular weight determination.

Cell culture laboratory

  • incubators, laminars, microscope, centrifuge
  • hypoxia chamber  - O2 control glove box for in vitro studies (Coy Life Products)

Cell imaging and analysis
